Production of Novel Alkali-thermo-tolerant Cellulase-poor Xylanases from Coprinopsis Cinerea Hk-1 Nfcci-2032

چکیده
Culture conditions of a newly isolated basidiomycetous strain were optimized for the enhanced production of extracellular alkali-thermotolerant cellulase-poor xylanase using wheat bran as the sole carbon source under solid state fermentation (SSF). SEM and ITS sequencing confirmed it as Coprinopsis cinerea HK-1 NFCCI-2032. Among various inexpensive agro-residues, wheat bran (carbon source) came up as the most potent enzyme inducer under SSF, and resulted in 54% higher xylanase activity compared to that in submerged fermentation mode. The strain grew well even at 47 oC. The highest xylanase (695.8 IU/mL) titer was recorded at a substrate:moisture ratio of 1:3 after 7 days of incubation at 37 C at pH 6.4 along with 0.541 IU/mL of poorly associated cellulase activity. The xylanase exhibited remarkable stability and retained 50% of its activity at pH 8.0 on incubation at 55 oC for 15 min and 78, 43, and 23% of its activity at temperatures 65, 75 and 85 C, respectively, demonstrating an approximately 50% alkali-thermo-tolerant nature, which is suitable for biobleaching.

Cellulose-degrading enzymes from Aspergillus terreus D34 and enzymatic saccharification of mild-alkali and dilute-acid pretreated lignocellulosic biomass residues
Background: Production of cellulose-degrading enzymes from Aspergillus terreus D34 using different growth substrates was studied under solid-state cultivation. We have tested two lignocellulosic biomass residues viz., rice straw (RS) and sugarcane bagasse (BG), both separately and in combinations, and crystalline cellulose as a sole source of carbon for cellulase production. We also demonstrate...

The importance of recycling and utilization of recycled fiber has recently been recognized because of concerns about environmental issues and economic aspects. In spite of several advantages offered by paper recycling such as wood savings through substitution of virgin pulp and reduction in chemical usage, recycled pulp also contains plenty of fines having high relative surface area and hig...

The production, optimisation and partial characterisation of xylanases from newly isolated wild strains of Coprinellus disseminatus was performed in solid-state fermentation. Strains SH-1 and SH-2 showed high xylanase (727.78 and 227.99 IU/mL) with very low CMCase (0.925 and 0.660 IU/mL) and laccase (0.640 and 0.742 U/mL) activities at incubation time seven days, 37 degrees C and initial pH 6.4...

Because of the high energy consumption for fermentor cooling, the isolation of thermo-tolerant Acetobacter strains for vinegar production has a high priority. The aims of this study were the isolation and identification of a high ethanol-thermo-tolerant Acetobacter spp. from grapes as well as the optimization of conditions for increasing the acetic acid production. The grape e...
